Mi piace il tuo sfidare i mezzi che hai a disposizione e così hai già ottenuta una risposta: meglio inversione del grandangolo. Lo scatto con unico semino è sicuramente più nitido ed il rapporto d'ingrandimento ottenuto mi sembra anche maggiore di quello di questo scatto.
Piacerebbe anche a me conoscere la tecnica di illuminazione che risulta omogenea ed immagino l'obiettivo vicinissimo al soggetto ripreso
Mi viene quindi la curiosità dei risultati ottenibili con scatto singolo, anziché una sequenza di ben 78 scatti in focus stacking, i semini infatti, non hanno enorme spessore. Mi chiedo se sia impossibile avere a fuoco seme e frutto con unico scatto e diaframma molto chiuso.
Nel mio scatto dedicato all'Ederina dei muri, ho usato, per la prima volta, due tubi di prolunga e la pdf non si è rivelata limitatissima come temevo.

Sì, sì, con RR superiori a 2 a mio modesti avviso stravince l'inversione, per nitidezza, ingrandimento e agilità dell'attrezzatura.
Per l'illuminazione utilizzo un anello a led da 6 o da 10 pollici, dipende dalle dimensioni del soggetto. Non sono di quelli che si collegano all'obiettivo, ma sono di quelli usb, con un proprio treppiede che su un noto shop online vengono proposti per illuminare il volto per i selfie o per il trucco delle donne... io li trovo comodi per fare macro 
Spesa 10-20 euro, lo alimento con un powerbank ed ho modificato un treppiedi per posizionarlo.
Personalmente in macro scarto l'ipotesi dello scatto singolo, oltre un certo punto noto una progressiva perdita di nitidezza e comunque la pdc non mi basta mai.
Sergio, per curiosità, ricordi che RR hai raggiunto con l'ederina?

A volte però qualche soggetto avrebbe giovato di un ingrandimento maggiore. Da qui il dubbio se croppare l'immagine scattata con il 100mm o acquistare i tubi di prolunga. Deciso per questa seconda strada e provare.
L'ederina ha goduto della prima serie di scatti di sperimentazione.
Non saprei dirti che RR abbia raggiunto con i due tubi di prolunga. Il fiore reale sarà profondo circa mezzo centimetro.

Considera che più allunghi e inverti, più la scena diventa buia.
Con tubi e inversione inoltre si perde la messa a fuoco all'infinito, problema che non riscontro con il moltiplicatore, ma purtroppo questo costa molto più di un kit di tubi, quindi se ne senti il bisogno poche volte in effetti ti conviene provare con i tubi.
